Overview

FOXO4-DRI is a synthetic D-retro-inverso peptide (built from D-amino acids in reversed order to resist breakdown) that blocks the FOXO4-p53 interaction senescent cells rely on to survive, pushing them into apoptosis. It was introduced in a 2017 Cell study in which it restored fitness, fur density and kidney function in fast-aging and naturally aged mice; all later work is also cell or animal research, and one mouse study found senolytic clearance with FOXO4-DRI worsened pulmonary hypertension. No human trial has been published. It is not FDA-evaluated and is sold for laboratory research only; nothing here is medical or dosing advice.
